產(chǎn)品介紹 P19ARF Capable of inducing cell cycle arrest in G1 and G2 phases. Acts as a tumor suppressor. Binds to MDM2 and blocks its nucleocytoplasmic shuttling by sequestering it in the nucleolus. This inhibits the oncogenic action of MDM2 by blocking MDM2-induced degradation of p53 and enhancing p53-dependent transactivation and apoptosis. Also induces G2 arrest and apoptosis in a p53-independent manner by preventing the activation of cyclin B1/CDC2 complexes. Binds to BCL6 and down-regulates BCL6-induced transcriptional repression. Binds to E2F1 and MYC and blocks their transcriptional activator activity but has no effect on MYC transcriptional repression. Binds to TOP1/TOPOI and stimulates its activity. This complex binds to rRNA gene promoters and may play a role in rRNA transcription and/or maturation. Interacts with NPM1/B23 and promotes its polyubiquitination and degradation, thus inhibiting rRNA processing. Interacts with UBE2I/UBC9 and enhances sumoylation of a number of its binding partners including MDM2 and E2F1. Binds to HUWE1 and represses its ubiquitin ligase activity. May play a role in controlling cell proliferation and apoptosis during mammary gland development.

p19ARF是抑癌基因,通過干擾p53的功能,導(dǎo)致細胞周期調(diào)節(jié)紊亂和細胞凋亡障礙,使細胞發(fā)生癌變,與多種腫瘤的形成有關(guān)。
目前對p19ARF的研究雖然取得了很大的進展,但是還存在一些亟待解決的問題,如:p19ARF蛋白的表達調(diào)控還不是十分清楚;如何正確地認(rèn)識并理解p19ARF的作用機制,p19ARF對腫瘤發(fā)生的研究有重要的理論意義;目前的許多證據(jù)大部分來源于動物實驗,p19ARF與人類腫瘤的關(guān)系還需要進一步研究。
